Optimisation of metabolism of Bacillus methanolicus to prevent CO2 loss during methanol-based growth

Research area and project description:
You will be part of project CO2MOS: CO2 Metabolism Optimisation Strategy to prevent CO2 loss during methanol assimilation. In this project a methylotrophic bacterium, Bacillus methanolicus, will be used as a model organism. B. methanolicus is a next candidate to become industrial workhorse for methanol-based production of value-added compounds. It is able to grow on methanol as sole carbon and energy source. However, it was previously observed that large proportion of carbon is lost in form of CO2 during methanol-based growth. In CO2MOS, we will identify the major contributors to CO2 loss in B. methanolicus using genome scale model of its metabolism and we will implement necessary changes in its genome to prevent it. This way, we will create a microbial strain that can be used for establishing efficient methanol-based bioproduction processes, solving an urgent issue of raw materials scarcity. the pathways contributing to carbon loss, and creation of respective deletion strains. The strain characterization will include metabolic flux analysis in collaboration international partner and evaluation of the feasibility for production of value-added compounds.

Qualifications and specific competences:
The ideal candidate has initiated or completed a relevant Masters’ degree in biotechnology, microbiology, biological engineering, molecular biology or other relevant subjects.

We are looking for a motivated PhD candidate interested in metabolic engineering. The project combines use of GSM for strain design, strain engineering and advanced strain characterisation including batch and fed-batch fermentations and metabolic flux analysis.

The preferred candidate meets following criteria:

  • experimental experience in molecular biology work
  • microbial cultivation techniques
  • analytical techniques for liquids
  • excellence in both written and spoken English
Parts of the experimental activities will be performed during an abroad research stay.
You will be working in the research group “Microbial Synthetic Biology” as part of the Dept. of Biological & Chemical Engineering. As a group member you will be expected to be a team player, where you contribute to student co-supervision, teaching and day-to-day work of the group.
